AMEX Travel Question - Leveraging Points for Flights (JFK-> BPS)

I'm trying to fly from JFK to BPS using AMEX POINTS via Delta Flight, I want to fly premium class, what is the best strategy in terms of transferring, leveraging, and for delta at times to that block out some flights from using points?
